Mayamohini follows Balakrishnan, a young heir raised by his uncle after his parents die in a plane crash, whose family fortune depends on him marrying a woman born under a specific star. When his new wife Maya abruptly leaves him, he hires a woman named Mayamohini to pose as his spouse to hide the truth from his uncle, only to discover that Mayamohini is actually a man, Mohanakrishnan, being sought by the police. Their partnership leads them to Mumbai in search of Mohanakrishnan's missing father, uncovering a web of deception tied to a wealthy businessman and his associates.
